Long-Term Potentiation: The Real Science Behind the Brain's 'Save Button'
The process supplement ads call a save button is one of neuroscience's great discoveries. Here is how it works, how it is measured, and what it does not prove.
What long-term potentiation is
Long-term potentiation (LTP) is a lasting increase in the strength of a synapse after it has been activated in a particular pattern. When a sending neuron repeatedly helps fire a receiving neuron, the connection between them becomes more effective, for hours, days or longer. First described in the rabbit hippocampus in 1973, it has become one of the best-studied processes in neuroscience and a leading cellular model of how memories are stored[1].
How it works, in four steps
- Glutamate released by the sending neuron reaches NMDA receptors on the receiving neuron.
- Coincidence detection: NMDA receptors open only if the receiving neuron is already active, so they register "these two fired together".
- Calcium entry switches on enzymes that insert more AMPA receptors, strengthening the synapse within minutes (early LTP).
- Gene expression: for lasting change, signals reach the nucleus, where CREB turns on genes, and BDNF and new proteins build new connection points (late LTP).
Where sleep fits
The "save" in everyday life happens largely offline. During deep sleep, the hippocampus replays the day's patterns and the cortex gradually takes them up, a process called systems consolidation[2]. That is why short sleep feels like poor memory, and why sleep is one of the most reliable ways to protect it.
Measuring it in people
LTP itself is measured in brain tissue. In living people, transcranial magnetic stimulation can induce a related "LTP-like" change in cortical excitability. In people with Alzheimer's disease that response is weaker, as a 2016 study from Rome showed[3]. This is research into disease mechanisms; it does not show that any product can restore the response.
How marketing uses it
Supplement presentations, including Mindoxa's, call LTP the brain's "save button" and assign ingredients to its steps: Cordyceps for the signal, Reishi for the CREB and BDNF step, Lion's Mane for structure. The underlying studies are real but mostly cellular or animal[5][6]. Meanwhile, the interventions with human evidence for the hippocampus are humbler: in older adults, a year of brisk walking enlarged it and improved memory[4].
Quick answers
Is the brain's 'save button' real?
The process it refers to, long-term potentiation, is real and well established. The 'save button' label and the idea that supplements restore it are marketing.
What strengthens memory formation most reliably?
Sleep, attention at the moment of learning, and regular aerobic exercise have the strongest evidence.
